摘要
為了將抗鹽壓力基因從紅樹林植物中脫離出來，組建了一個紅海欖根的互補DNA庫，並用基於聚合酶鏈反應（PRC）的抑制消減雜交法（SSH）來帥選壓力相關的基因。紅樹幼苗在兩種條件下生長了8個月：一種是讓其接觸含鹽量為3%的水，另一種是讓其接觸淡水。將接觸含3%鹽水的幼苗的互補DNA用作檢驗品，將接觸淡水的幼芽用作驅動品。我們從SSH庫中隔離出240個（DNA）並用正調節表現度序列標簽作了標注。在這些正調節表達序列標簽中，對48個獨特克隆作了推定識別，並將其歸入10個功能類別，如細胞救援與防護，次級代謝，蛋白質合成，新陳代謝等。從各類別選出15個基因，用實時逆轉錄酶-聚合酶鏈反應（RT-PCR）對其活躍度進行研究。記錄結果確定這15個基因中的13個的活躍程度有了顯著提高，這說明這些基因對此種植物的耐鹽性起了作用。在它們中間，首次從紅樹植物中識別出了參與類異戊二烯生物合成的兩個轉錄因子及多個基因具有耐鹽性。論述了將紅樹植物長期接觸鹽壓力的情況下這些基因活躍度提高在生理學上的意義。
